Reagent for serological diagnosis of Brucellosis, smooth type, for veterinary use

Composition:

Bacterial mass of Brucella abortus S1119-3 8% of cell concentration

Sterile saline solution with 0.5% of phenol               q.s. 10 liters

Bengal Rose Colorant solution 1% (w/v)               285 mL

Sodium hydroxide 20 g

Lactic acid 90 mL

Indications for use:

Reagent for serological diagnosis of Brucellosis, smooth type, for veterinary use.

Target species:

Bovine, buffalo or swine serum, or other animals specifically requested by Ministries may be used

to make the diagnosis of Brucellosis using this reagent.

Route of administration and dosage:

Not directly applied to animals.

Serological diagnostic technique using Pink Bengal reagent:

  • Balance serum and reagent at room temperature for at least 30 minutes. In case the serum is frozen, this time will be longer.
  • Homogenize the serum before running the test. Samples must be individually identified.
  • Use a micropipette or syringe or other system, take 30 μL of serum and place it on the glass plate inside the designated area. Change of type or material between serum. Stir the plate gently.
  • Place 30 μL of Pink Bengal reagent next to each drop of serum within the enclosed area of the plate.
  • Mix each drop of serum with the drop of reagent next to it, shaking the plate with circular movements so as to obtain a circle of approximately 2 cm of diameter, always respecting the limit line of each area.
  • Shake the plate with oscillatory movements with a frequency of approximately 30 movements per minute, so that the mixture flows slowly within each delimited circle. The plate should be shaken continuously for 4 minutes.

      Place the plate under indirect light and read.

          Record the results. Agglutination reactions occurring after 4 minutes should not be considered.

             Interpretation of results:

* Presence of lumps: reagent

* Absence of lumps: Not reactive.
