The tuberculin is a product of guaranteed use for diagnosis of tuberculosis. It is universally used in programs for control and eradication of the cattle disease.<\/strong><\/p>\n

The purified protein derivative (PPD) is the purified protein produced from cultures of inactivated AN-5 strain of Mycobacterium bovis<\/em><\/strong> at a concentration of 1 mg\/mL. It is used as an antigen for the skin test to identify animals suffering from tuberculosis.<\/p>\n

The tuberculin test has proven to be the main tool for the detection of tuberculosis worldwide and is the only officially approved method for the diagnosis of the disease.<\/p>\n

Intradermal tuberculin test in cattle<\/strong><\/p>\n

The caudal-fold test<\/strong> involves the intradermal injection of 0.1 mL of bovine PPD tuberculin in the middle third of the internal caudal-fold, about 6 cm from the base of the tail and in the centre of the fold, after cleaning the region with non-irritating substances. The needle must be inserted completely by intradermal route, and partially withdrawn to inject the tuberculin. In a well-applied injection, a papule will appear on the inoculated site.<\/p>\n

Reactions must be read 72 hours after the injection ( \/- 6 hours), slightly lifting the tail to stretch the fold. The site of application must be evaluated by observation and palpation, comparing the injected fold skin thickness with the opposite fold, which will serve as a witness. The fold must be palpated with the thumb and forefinger to check for thickening and to measure it exactly using a calibre. If unable to do so due to weather or other causes, the reading can be made until 24 hours later.<\/p>\n

The simple cervical test <\/strong>involves intradermal inoculation of 0.1 mL of bovine PPD at a concentration of 1 mg\/mL in the middle third of the neck. Hair is cut with scissor or machine at the site of injection in an area of 5 to 6 cm2<\/sup>, skin thickness is measured with a calibre and data is recorded. Readings must be done 72 hours after the injection ( \/- 6 hours). The skin thickness measurement must be repeated and compared with the measurement made previous to the injection.<\/p>\n

For purposes of sanitation all females and males over 6 months of age should undergo diagnostic testing.<\/p>\n

Intradermal tuberculin test in pigs<\/strong><\/p>\n

The test involves the intradermal injection of 0.1 mL of bovine PPD (simultaneous injection of avian PPD may be carried out because of the multiple susceptibility of this species) at the base of the right and left ear, if using both tuberculins. Readings are carried out after 48 hours.<\/p>\n
